change: have to restart all data collection.

we are changing from e. coli MM294 to strain K12. fml. but nvm. shldn't take more than 2.5 weeks to generate this result.

found a way to add the catalyst. according to boss, can use methylene blue in broth side.

den we analyse kinetics. shld b quite scientifically sound.

but main analysis is still on glu conc, catalyst conc, fruit peel extract conc.

detail:

bacteria cell conc:

absorbance read at 600nm, blank of nb. check absorbance of bacteria in broth for 1.000 against blank, if not adjust accordingly
3% glu nb
0.061V stable. repeating next mon

change: carbon rod fully submerged in bacterial broth.

1% glu, nb 1l, CuSO4.
stable: 0.060V as of 24h

Monday, November 9, 2009

3% glu, nb, bacteria 60ml, CuSO4 1%
0.045V stable.

Thursday, November 5, 2009

making broth:
  1. weigh in mass of nb for 1l
  2. weigh in % of glu
  3. put in 1000ml water
  4. autoclave
  5. 50ml bacteria

bacteria broth making:

  1. 50ml broth
  2. ? loops of bacteria
  3. incubate at 30degC overnight

cathode solution: bacteria. electrode: carbon rod
anode solution: copper(II) sulfate. electrode: copper piece.

1st run:
5% glu, nb, bacteria 1l, CuSO4 1%.
0.306V max stable.

2nd run:
7% glu, nb, bacteria 1l, CuSO4 1%
0.045V stable.
